Drug intelligence / Profile preview
An anti-CSF1 neutralizing antibody is a therapeutic monoclonal antibody designed to target and neutralize macrophage colony-stimulating factor 1 (CSF1, also known as M-CSF). CSF1 is a key cytokine that regulates the survival, proliferation, differentiation, and chemotaxis of macrophages and monocytes via its receptor, CSF1R. In the tumor microenvironment, tumor-derived CSF1 recruits and polarizes tumor-associated macrophages (TAMs) into an immunosuppressive M2-like phenotype, promoting tumor growth, angiogenesis, and metastasis. By binding to CSF1 and blocking its interaction with CSF1R, anti-CSF1 neutralizing antibodies deplete or reprogram TAMs, alleviating local immunosuppression and enhancing antitumor immune responses, particularly when combined with chemotherapy or immune checkpoint inhibitors. Clinical-stage candidates in this class include lacnotuzumab (MCS110) and PD-0360324.
