Overview

Anti-CSF1-R antibodies are a class of monoclonal antibodies designed to target and inhibit the Colony Stimulating Factor 1 Receptor (CSF-1R), also known as M-CSF receptor or CD115. CSF-1R is a receptor tyrosine kinase that plays a critical role in the differentiation, proliferation, and survival of cells in the monocytic lineage, particularly tumor-associated macrophages (TAMs). By blocking the binding of ligands CSF-1 and IL-34, these antibodies aim to deplete immunosuppressive TAMs within the tumor microenvironment, thereby enhancing anti-tumor immune responses. This modality is being investigated across various oncology indications, including solid tumors and tenosynovial giant cell tumors (TGCT), often in combination with immune checkpoint inhibitors. Multiple distinct agents have been developed in this class, including emactuzumab, cabiralizumab, and axatilimab.
