Drug intelligence / Profile preview
Anti-CTHRC1 monoclonal antibody refers to a class of monoclonal antibodies targeting collagen triple helix repeat-containing protein 1 (CTHRC1), a secreted glycoprotein involved in extracellular matrix remodeling, vascular remodeling, and fibroblast activation. In the context of cardiovascular disease, CTHRC1 is upregulated in response to cardiac stress and secreted by activated cardiac fibroblasts. It binds to the ADAM17 receptor on cardiomyocytes, activating the NF-κB pathway and the NLRP3 inflammasome, which drives myocardial pyroptosis, cardiac fibrosis, and diastolic/systolic dysfunction. Treatment with anti-CTHRC1 monoclonal antibodies has been shown in preclinical models to inhibit this signaling cascade, reducing stress-induced cardiac fibrosis and improving cardiac function. Additionally, CTHRC1 is overexpressed in various solid tumors (such as pancreatic, ovarian, and colorectal cancers) where it promotes tumor progression, metastasis, and tumor-associated fibrosis; therapeutic antibodies targeting CTHRC1 (such as Prestige Biopharma's PBP1710) are also under development for oncology and broader fibrotic indications.
