Anti-CTLA-4 (clone 9H10) is a Syrian hamster monoclonal antibody that specifically targets murine Cytotoxic T-Lymphocyte-Associated protein 4 (CTLA-4, also known as CD152). CTLA-4 is a critical immune checkpoint molecule expressed on the surface of activated T cells and regulatory T cells (Tregs). It functions as a negative regulator of T cell immune responses by outcompeting the costimulatory receptor CD28 for binding to its ligands, CD80 (B7-1) and CD86 (B7-2), on antigen-presenting cells. Clone 9H10 is a widely utilized research tool in preclinical oncology and immunology studies to achieve CTLA-4 blockade. By preventing CTLA-4-mediated inhibition, the antibody enhances T cell proliferation, effector function, and antitumor immunity. In recent research, clone 9H10 has been used in mouse models (such as B16F10 melanoma and MC38 colorectal cancer) to investigate the therapeutic window of checkpoint inhibitors and the management of immune-related adverse events (irAEs) using sequential CTLA-4 Ig treatment.
