Anti-CTLA-4 IgG2a is a monoclonal antibody targeting cytotoxic T lymphocyte-associated antigen 4 (CTLA-4), an immune checkpoint receptor that inhibits T cell activation. The IgG2a isotype is used primarily in preclinical mouse models and is characterized by an Fc region that confers strong antibody-dependent cellular cytotoxicity (ADCC) and the ability to deplete intratumoral regulatory T cells (Tregs) via engagement of Fc gamma receptors on macrophages, enhancing the Teffector:Treg cell ratio and promoting antitumor immune responses[3][4][5]. Its mechanism includes CTLA-4 blockade, leading to enhanced T cell activity and selective Treg depletion within tumors, with activity highly dependent on Fc-FcγR interactions[3][4][5]. It is considered a tool compound for experimental cancer immunotherapy.
