The anti-CXCR1 nanobody is a high-affinity nanobody designed to block CXCR1, a chemokine receptor expressed on tumor-infiltrating natural killer (NK) cells. By targeting CXCR1, this nanobody aims to prevent NK cell exhaustion and improve their infiltration into tumors, thereby enhancing NK cell-mediated anti-tumor immunity. It has demonstrated superior tumor penetration compared to conventional antibodies and exhibited favorable safety profiles in models. Preclinical studies suggest it can outperform PD-1 inhibition in controlling tumor growth and prolonging survival, particularly in hepatocellular carcinoma (HCC) models, by modulating chemokine signaling.
