Anti-CXCR5 CAR T-cell therapy is an investigational chimeric antigen receptor (CAR) T-cell immunotherapy jointly developed by the Max Delbrück Center and Charité – Universitätsmedizin Berlin. The therapy involves engineering a patient's T-cells to express a CAR consisting of a novel single-chain variable fragment (scFv) targeting C-X-C chemokine receptor type 5 (CXCR5), coupled with CD28 costimulatory and CD3z signaling domains. CXCR5 is a G protein-coupled receptor restricted to mature B cells and follicular helper T (Tfh) cells, making it a stable target for B-cell non-Hodgkin's lymphomas (B-NHL) and Tfh-derived T-cell lymphomas. A distinctive feature of this therapy is its dual mechanism of action, which simultaneously targets malignant lymphoma cells and the tumor-promoting Tfh cells within the microenvironment. The technology has been licensed to the spin-off company CARTemis Therapeutics, and clinical development is supported by the German Federal Ministry of Education and Research (BMBF).
