Anti-CXCR7 antibodies are a class of experimental monoclonal antibodies that target the Atypical Chemokine Receptor 3 (ACKR3), formerly known as CXCR7. Unlike classical G protein-coupled receptors, CXCR7 primarily signals through the β-arrestin pathway and acts as a high-affinity scavenger for the chemokines CXCL11 and CXCL12 (SDF-1). By binding to CXCR7, these antibodies can block the sequestration of chemokines, thereby modulating the CXCR4/CXCR7/CXCL12 signaling axis which is frequently dysregulated in various pathologies. Preclinical research has explored these antibodies for their potential to inhibit tumor cell proliferation, migration, and metastasis in cancers such as renal cell carcinoma, prostate cancer, and glioma, as well as for therapeutic applications in inflammatory diseases, multiple sclerosis, and HIV.
