Anti-DC-HIL monoclonal antibody is a research-stage biologic targeting DC-HIL (also known as Glycoprotein Nonmetastatic Melanoma Protein B, GPNMB), a T-cell-inhibitory receptor. DC-HIL is expressed on monocytic myeloid-derived suppressor cells (MDSCs) and certain tumor cells, where it mediates immunosuppression by binding to syndecan-4 on T cells. By blocking this interaction, the monoclonal antibody aims to restore T-cell activity and enhance anti-tumor immune responses. Preclinical studies, primarily conducted at UT Southwestern Medical Center, have demonstrated that anti-DC-HIL mAb can reverse MDSC-mediated T-cell suppression, attenuate tumor growth, and reduce lung metastasis in models of melanoma and colorectal cancer.
