Anti-DDR1 antibody refers to a class of investigational therapeutic monoclonal antibodies and antibody-drug conjugates (ADCs) designed to target discoidin domain receptor 1 (DDR1, also known as CD167a). DDR1 is a collagen-activated receptor tyrosine kinase that is highly expressed in various solid tumors, including breast, non-small cell lung, colorectal, and gastric cancers. In the tumor microenvironment, DDR1 binds to collagen fibers and promotes their alignment, creating a physical barrier that excludes cytotoxic T cells from infiltrating the tumor core. By blocking the interaction between DDR1 and collagen, anti-DDR1 antibodies disrupt this collagen barrier, allowing immune cells to access and attack the tumor. The most advanced candidate in this class is PRTH-101, which is being evaluated in Phase I clinical trials as a monotherapy and in combination with immune checkpoint inhibitors.
