Anti-DKK-1 antibodies are a class of monoclonal antibodies designed to target and neutralize Dickkopf-related protein 1 (DKK-1), a secreted protein that acts as a potent antagonist of the canonical Wnt/β-catenin signaling pathway. DKK-1 is frequently overexpressed in various malignancies, including multiple myeloma, hepatocellular carcinoma, and prostate cancer, where it contributes to osteolytic bone disease by inhibiting osteoblast differentiation and promotes an immunosuppressive tumor microenvironment. By blocking the interaction between DKK-1 and its receptors (LRP5/6 and Kremen), these antibodies aim to restore Wnt signaling, thereby promoting bone formation and enhancing anti-tumor immune responses. Clinical-stage candidates in this class include BHQ880, developed by Novartis for multiple myeloma, and DKN-01, developed by Leap Therapeutics for gastroesophageal and gynecologic cancers.
