Anti-DKK1 refers to a class of monoclonal antibodies designed to target and neutralize Dickkopf-related protein 1 (DKK1), a secreted protein that acts as a potent antagonist of the canonical Wnt/β-catenin signaling pathway. By binding to DKK1, these antibodies prevent its interaction with the LRP5/6 receptors, thereby restoring Wnt signaling. This pathway is a critical regulator of bone homeostasis, promoting osteoblast differentiation and bone formation while inhibiting bone resorption. Consequently, anti-DKK1 agents are primarily investigated for their bone-anabolic potential in conditions such as multiple myeloma-induced bone disease and osteoporosis. Furthermore, because DKK1 is overexpressed in various malignancies and contributes to an immunosuppressive tumor microenvironment, anti-DKK1 antibodies (such as DKN-01) are also being evaluated as oncology therapeutics to enhance anti-tumor immune responses and inhibit tumor growth.
