anti-DR5 CAR-T refers to chimeric antigen receptor T cell therapy engineered to specifically target death receptor 5 (DR5), a member of the tumor necrosis factor receptor superfamily predominantly expressed on tumor cells. The therapy involves isolating patient T cells, genetically modifying them with a CAR construct that recognizes DR5, and expanding these cells for reinfusion. Once infused, the CAR-T cells bind DR5 on tumor cells, leading to tumor cell apoptosis via extrinsic death signaling. DR5 activation triggers assembly of the death-inducing signaling complex (DISC), which recruits FADD and activates caspase-8, culminating in tumor cell death. This therapy is under investigation primarily for cancer indications, especially solid tumors that overexpress DR5. Developers and manufacturers may vary based on academic and clinical trial sponsorship.
