Overview

anti-DR5 IgG refers to a class of monoclonal antibodies of the immunoglobulin G (IgG) isotype that target Death Receptor 5 (DR5), also known as TRAIL receptor 2 (TRAIL-R2). These antibodies are designed to mimic the action of the Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand (TRAIL) by binding to DR5 on the surface of tumor cells and triggering the extrinsic apoptotic pathway. While bivalent IgG antibodies can bind DR5, they often require additional cross-linking (e.g., via Fc gamma receptors in the tumor microenvironment) to achieve the high-order receptor clustering necessary for potent caspase activation and cell death. Clinical candidates in this class, such as mapatumumab and conatumumab, have historically faced challenges in demonstrating robust clinical efficacy as monotherapies, leading to the development of next-generation multivalent formats like IgMs or multi-specific antibodies.
