Overview

An anti-EGF antibody is a monoclonal antibody designed to specifically bind and neutralize the Epidermal Growth Factor (EGF) ligand. By sequestering EGF, the antibody prevents it from interacting with its cognate receptor, the Epidermal Growth Factor Receptor (EGFR/HER1). This neutralization inhibits the activation of downstream signaling cascades, such as the Ras/MAPK and PI3K/Akt pathways, which are critical for cell proliferation, survival, and motility. In oncology research, anti-EGF antibodies are utilized to disrupt paracrine signaling loops between tumor cells and the microenvironment, such as the stimulation of glioma cell invasion by microglia. While most clinical focus in the ErbB pathway has been on receptor-targeting agents (e.g., cetuximab or panitumumab), anti-EGF antibodies serve as important tools for studying ligand-dependent mechanisms of tumor progression and are explored in preclinical models to evaluate the therapeutic potential of ligand sequestration.
