Overview

An anti-EGFR blocking antibody is a targeted biologic therapy designed to bind specifically to the extracellular domain of the epidermal growth factor receptor (EGFR). By binding to EGFR, these antibodies competitively inhibit the binding of natural ligands, such as epidermal growth factor (EGF) and transforming growth factor-alpha (TGF-alpha), thereby preventing receptor dimerization, autophosphorylation, and downstream signaling cascades (such as the RAS/MAPK and PI3K/Akt pathways) that drive cell proliferation, survival, and metastasis. Approved monoclonal antibodies within this class, such as cetuximab and panitumumab, are widely utilized in the treatment of EGFR-expressing malignancies, including metastatic colorectal cancer and squamous cell carcinoma of the head and neck.
