Anti-EGFR CAR-T cells are an adoptive cellular immunotherapy consisting of T cells engineered to express a chimeric antigen receptor (CAR) specific for the epidermal growth factor receptor (EGFR). EGFR is a transmembrane protein frequently overexpressed in various solid tumors, including glioblastoma, bladder cancer, and lung cancer, making it a prominent target for CAR-T development. Upon binding to EGFR on the surface of tumor cells, these CAR-T cells are activated, leading to the release of cytotoxic granules (such as granzyme B) and pro-inflammatory cytokines (like IFN-γ and TNF-α), ultimately resulting in tumor cell lysis. Research is ongoing to address challenges such as antigenic heterogeneity and the immunosuppressive tumor microenvironment, sometimes through the development of multi-targeted (e.g., bicistronic EGFR/IL13Rα2) constructs.
