Drug intelligence / Profile preview
anti-EGFR-ILs-dox is a targeted nanocontainer drug developed for the treatment of EGFR-overexpressing malignancies. It consists of pegylated liposomes containing doxorubicin, with anti-EGFR antibody fragments (derived from cetuximab) covalently inserted into the lipid bilayer. This immunoliposomal delivery system is designed to enhance the therapeutic index of doxorubicin by increasing drug concentration at the tumor site and facilitating intracellular delivery via receptor-mediated endocytosis, potentially overcoming multidrug resistance. While it showed a manageable safety profile in Phase I and II trials, a Phase II study in triple-negative breast cancer (TNBC) failed to meet its primary endpoint of progression-free survival, leading to the discontinuation of development for that specific indication. It has also been investigated in glioblastoma.
