Anti-EGFR monoclonal antibody 528 (clone 528) is a mouse monoclonal antibody directed against the extracellular domain of the epidermal growth factor receptor (EGFR). It specifically binds to domain III of EGFR at an epitope that is distinct from, but partially overlaps with, the cetuximab binding site. Primarily utilized as a research reagent, antibody 528 has been extensively used in studies of EGFR signaling, receptor-ligand interactions, and as a building block for engineering novel therapeutics, such as bispecific T-cell engagers. It effectively competes with natural ligands like EGF and TGF-alpha, thereby inhibiting receptor autophosphorylation and downstream signaling cascades that drive tumor cell growth and metastasis. While it has demonstrated efficacy in various preclinical cancer models, including prostate and breast cancer cell lines, it is not currently in clinical development as a standalone therapeutic.
