Drug intelligence / Profile preview
EGFR targeting drug conjugate (also known as Anti-EGFR Nanofitin D8-MMAE) is a preclinical-stage small antibody-drug-conjugate-like molecule (specifically a Nanofitin-drug conjugate) being developed by Affilogic. It consists of a monovalent anti-EGFR Nanofitin (specifically the D8 clone, a 7 kDa alternative protein scaffold) conjugated to the cytotoxic payload monomethyl auristatin E (MMAE). By targeting the epidermal growth factor receptor (EGFR) overexpressed on tumor cells, the conjugate is internalized and releases MMAE to inhibit tubulin polymerization, leading to cell cycle arrest and apoptosis. In preclinical in vivo xenograft models of EGFR-positive solid tumors, the conjugate demonstrated deep tumor penetration, rapid systemic clearance, and significant tumor growth inhibition.
