Overview

anti-EGFRvIII ADC 5 is an experimental antibody-drug conjugate (ADC) targeting the Epidermal Growth Factor Receptor variant III (EGFRvIII), specifically developed for the treatment of glioblastoma multiforme (GBM). It is a homogeneous conjugate with a drug-to-antibody ratio (DAR) of 4.0, produced using site-specific conjugation technology (AbClick). The drug is designed to overcome the limitations of conventional heterogeneous ADCs, which often contain highly loaded species that fail to penetrate the blood-brain barrier (BBB). By maintaining homogeneity and an optimal payload loading rate, anti-EGFRvIII ADC 5 demonstrates enhanced delivery of its cytotoxic payload, monomethyl auristatin E (MMAE), to brain tumor tissues, resulting in improved antitumor efficacy and survival in preclinical orthotopic models.
