Overview

Anti-endoglin siRNA is a small interfering RNA (siRNA) designed to silence the expression of endoglin (ENG, CD105), a type I transmembrane glycoprotein and co-receptor for the TGF-beta superfamily. Endoglin is highly expressed on the surface of proliferating vascular endothelial cells and certain chemoresistant tumor cells, making it a dual target for anti-angiogenic and cell-specific therapy. By reducing endoglin levels, this siRNA induces double-stranded DNA damage and suppresses the expression of key DNA repair genes, including BARD1 and BRCA1, thereby enhancing the efficacy of platinum-based chemotherapies like carboplatin and cisplatin. In preclinical research, it has been formulated using chitosan-encapsulated nanoparticles for systemic delivery in orthotopic ovarian cancer models.
