Overview

The anti-ENPP1 nanobody (specifically clone B7) is a therapeutic nanobody developed by the Moffitt Cancer Center that targets ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1). ENPP1 is an ectonucleotidase that degrades extracellular 2'3'-cGAMP, thereby suppressing cGAS-STING-mediated antitumor immunity. By blocking ENPP1, the nanobody prevents cGAMP degradation, which restores STING activation, enhances T-cell infiltration and effector function, and reverses myeloid-mediated immunosuppression in the tumor microenvironment. In preclinical models of homologous recombination-deficient (such as Brca1-deficient) breast cancer, therapeutic targeting with this nanobody has been shown to improve tumor control and synergize with PARP inhibitors and anti-PD-1 immunotherapy.
