Overview

Anti-EphA2 CAR-T cells is an investigational chimeric antigen receptor (CAR) T-cell therapy developed by Fuda Cancer Hospital in Guangzhou, China, for the treatment of EphA2-positive malignant glioma. The therapy involves the genetic modification of a patient's own T cells to express a CAR that specifically targets the Erythropoietin-producing hepatocellular carcinoma A2 (EphA2) receptor. EphA2 is a receptor tyrosine kinase that is highly overexpressed in approximately 90% of glioblastoma tissues but is found at low levels in normal brain tissue, making it a viable target for selective immunotherapy. Once infused, these engineered T cells are designed to recognize and bind to EphA2 on the surface of tumor cells, triggering T-cell activation and subsequent tumor cell lysis. A Phase 1/2 clinical trial (NCT02575261) was initiated to evaluate the safety and efficacy of this therapy in patients with recurrent and metastatic malignant glioma, but the study was later withdrawn, and the program has not progressed to later stages of development.
