Overview

Anti-EphA3 antibodies are a class of monoclonal antibodies designed to target the Erythropoietin-producing hepatocellular receptor A3 (EphA3), a member of the Eph receptor tyrosine kinase family. EphA3 is frequently overexpressed in various hematologic malignancies (such as acute myeloid leukemia and myelofibrosis) and solid tumors (including glioblastoma and head and neck cancers), while remaining largely absent in normal adult tissues. By binding to EphA3, these antibodies can induce direct tumor cell killing, disrupt the tumor microenvironment, or inhibit angiogenesis. The most clinically advanced candidate in this class was KB004 (ifabotuzumab), which reached Phase I/II trials. Other research-stage antibodies like Ea3Mab-20 are also being explored for diagnostic and therapeutic applications, including combination strategies with checkpoint inhibitors like CD47 blockades.
