anti-EphA3 antibody + temozolomide is a targeted nanomedicine combination consisting of the alkylating agent temozolomide (TMZ) conjugated to gold nanoparticles (GNPs) and functionalized with a targeting antibody against ephrin type-A receptor 3 (EphA3). Developed for the treatment of glioblastoma multiforme (GBM), particularly drug-resistant forms, this formulation is designed for intranasal administration to bypass the blood-brain barrier and directly reach the central nervous system. The anti-EphA3 antibody facilitates active targeting of glioma cells and glioma stem cells that overexpress the EphA3 receptor. Once internalized, temozolomide induces DNA damage and apoptosis. Additionally, the gold nanoparticle core enables auxiliary plasmonic photothermal therapy (PPTT) upon laser irradiation, which enhances the cytotoxic effect and helps reverse TMZ resistance by down-modulating the expression of O6-methylguanine-DNA methyltransferase (MGMT).
