anti-EphA3-TMZ16e-NPs is an experimental nanoparticle-based biologic formulation designed for the targeted treatment of glioblastoma (GBM), particularly drug-resistant forms. The formulation consists of nanoparticles loaded with temozolomide hexadecyl ester (TMZ16e), a highly lipophilic and membrane-permeable derivative of the alkylating agent temozolomide. The surface of these nanoparticles is modified with an anti-EphA3 antibody to specifically target the ephrin type-A receptor 3 (EphA3), which is highly expressed on glioma-initiating cells but has low expression in normal brain tissue. This targeted delivery system is intended for intranasal administration to bypass the blood-brain barrier, thereby enhancing drug concentration in the brain while reducing peripheral toxicity.
