Overview

Anti-factor IX monoclonal antibodies are a class of therapeutic biologics designed to bind to and inhibit the activity of coagulation factor IX (FIX) or its activated form (FIXa). By targeting the intrinsic pathway of the blood coagulation cascade, these antibodies serve as anticoagulants for the prevention and treatment of thromboembolic diseases. A specific research initiative at Beijing Children’s Hospital, Capital Medical University, involves identifying and characterizing natural human anti-FIX antibodies derived from pediatric hemophilia B patients who have developed inhibitors. This exploratory study utilizes single B-cell sequencing to discover antibodies with potent in vitro anticoagulant activity and evaluates their antithrombotic effects and safety profiles in humanized mouse models. The goal is to develop novel antithrombotic therapies that provide effective anticoagulation with a reduced risk of bleeding complications compared to traditional anticoagulants.
