Anti-Fas monoclonal antibody is a laboratory-generated antibody specific for the human Fas antigen (CD95), a cell surface death receptor associated with the tumor necrosis factor (TNF) receptor superfamily. Anti-Fas monoclonal antibodies can be classified as either agonist (e.g., CH11, E09) or antagonist (e.g., ZB4), with agonists binding to Fas and triggering apoptosis in target cells through Fas-mediated signaling. These antibodies selectively induce cell death in Fas-expressing cells by mimicking the effect of Fas ligand (FasL), leading to caspase activation and DNA fragmentation. They have been studied primarily in oncology, immunology, autoimmune diseases such as pemphigus and rheumatoid arthritis, and cell biology as research tools. Some forms like E09 are fully humanized; others such as CH11 are murine. Preclinical studies have reported cytotoxic activity toward HIV-infected cells, tumor cells (to a limited extent in human tumor lines), and potential immunomodulation by inducing apoptosis in activated immune cells[1][3][4][5][6][7].
