Overview

Anti-FcRL5 TCE refers to a class of bispecific T-cell engagers (TCEs) designed to treat multiple myeloma by targeting Fc receptor-like 5 (FcRL5), a surface protein highly expressed on malignant plasma cells. These agents are engineered with dual binding domains: one specific for FcRL5 on the tumor cell and another for the CD3 epsilon subunit of the T-cell receptor complex. By cross-linking these two cells, the TCE induces T-cell activation, secretion of cytotoxic granules (perforin and granzymes), and subsequent lysis of the myeloma cell, independent of MHC-I presentation. The most prominent clinical candidate in this class is cevostamab (BFCR4350A/RG6160), developed by Genentech and Roche, which has demonstrated clinical activity in patients with relapsed or refractory multiple myeloma, including those previously treated with BCMA-targeted therapies.
