Drug intelligence / Profile preview
An anti-FGFR1 blocking antibody is a monoclonal antibody designed to target and inhibit fibroblast growth factor receptor 1 (FGFR1). FGFR1 is a receptor tyrosine kinase that plays a critical role in cell proliferation, survival, and migration, and its aberrant activation is implicated in various cancers, fibrotic conditions, and inflammatory diseases like rheumatoid arthritis. By binding to the extracellular domain of FGFR1, the antibody blocks the binding of FGF ligands (such as FGF2), thereby preventing receptor dimerization, autophosphorylation, and the activation of downstream signaling cascades like the MAPK/ERK pathway. In oncology, FGFR1 blockade is being investigated to overcome resistance to immunotherapy and endocrine therapies, while in inflammatory diseases, it aims to suppress pathogenic fibroblast activation and cytokine production.
