Overview

Anti-fibulin-3 CAR-T cells are an experimental second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target EFEMP1 (also known as fibulin-3), a glycoprotein that is highly expressed in the extracellular matrix of glioblastoma (GBM) but absent in normal brain tissue. Developed by researchers at SUNY Upstate Medical University, these engineered T cells recognize the fibulin-3 present in the tumor's glycocalyx, leading to T-cell activation, proliferation, and the secretion of cytotoxic factors such as interferon-gamma, interleukin-2, and granzyme B. Preclinical studies have demonstrated that both local and intravenous administration of these CAR-T cells can significantly extend survival in intracranial and subcutaneous glioblastoma xenograft models, suggesting potential for treating the heterogeneous and immunosuppressive environment of GBM.
