Overview

Anti-fluorescein CAR T cell (also referred to as anti-FITC CAR T cell or AdFITC-CAR T cell) is an investigational, universal adaptor-based chimeric antigen receptor (CAR) T-cell therapy. Originally developed from research at Purdue University and Seattle Children's Therapeutics, and in development by Umoja Biopharma, this platform utilizes T cells engineered to express a CAR construct that recognizes the synthetic hapten fluorescein (FITC) with extremely high affinity. Rather than directly targeting a fixed tumor-associated antigen, these CAR T cells are redirected to cancer cells or tumor stroma via fluoresceinated bispecific adaptor molecules (such as the folate-fluorescein adapter UB-TT170). This modular design allows for precise control over CAR T-cell activation, dose titration, multi-antigen targeting to overcome tumor heterogeneity, and rapid termination of activity using free fluorescein to mitigate toxicities like cytokine release syndrome. The platform is currently being evaluated in Phase 1 clinical trials for the treatment of refractory or recurrent osteosarcoma.
