anti-folate receptor alpha-CAR4 is a fourth-generation chimeric antigen receptor (CAR) T-cell therapy designed to target Folate Receptor Alpha (FRα), a protein frequently overexpressed in solid tumors such as non-small cell lung cancer (NSCLC). The construct features a multi-domain signaling backbone consisting of CD28, 4-1BB (TNFRSF9), CD27, and the CD3 zeta chain. This fourth-generation design aims to enhance T-cell persistence and anti-tumor activity compared to earlier generations. In preclinical studies, it has demonstrated cytotoxicity against FRα-expressing lung cancer cell lines and served as a comparative baseline for fifth-generation CAR-T cells engineered with additional immunomodulatory features, such as checkpoint inhibitor secretion.
