anti-folate receptor alpha-CAR5 is an experimental fifth-generation chimeric antigen receptor (CAR) T-cell therapy designed to target Folate Receptor Alpha (FRα) in solid tumors, specifically non-small cell lung cancer (NSCLC). The construct is built upon a fourth-generation CAR backbone that incorporates CD28, 4-1BB, CD27, and CD3 zeta signaling domains. A distinguishing feature of this therapy is its ability to secrete a single-chain variable fragment (scFv) targeting programmed death-ligand 1 (PD-L1), which is derived from the monoclonal antibody atezolizumab. This dual-action design is intended to directly target FRα-expressing tumor cells while simultaneously neutralizing PD-L1-mediated immunosuppression within the tumor microenvironment, thereby enhancing the expansion, persistence, and cytotoxic efficacy of the CAR T cells.
