"Anti-FOLR1" refers to a class of therapeutic agents, including monoclonal antibodies, antibody-drug conjugates (ADCs), and T-cell engagers (TCEs), that target Folate Receptor Alpha (FOLR1). FOLR1 is a glycosylphosphatidylinositol (GPI)-anchored membrane protein that is highly expressed in several solid tumors—most notably ovarian, endometrial, and non-small cell lung cancers—while maintaining restricted expression in normal tissues, making it an attractive target for precision oncology. The context specifically highlights a new generation of FOLR1-targeting bispecific T-cell engagers developed by Amberstone Biosciences using their proprietary T-MATE (Tumor-Microenvironment Activated ThErapeutic) technology. These TCEs are engineered with pH-dependent CD3 antibodies that exhibit high potency in the acidic tumor microenvironment but minimal activity under normal physiological conditions, thereby potentially expanding the therapeutic index and reducing systemic toxicities like cytokine release syndrome. Preclinical studies have demonstrated significant tumor regression in xenograft models and a favorable safety profile in non-human primates.
