Overview

An anti-FPR1 blocking antibody is a monoclonal antibody designed to target and inhibit the Formyl Peptide Receptor 1 (FPR1). FPR1 is a G protein-coupled receptor primarily expressed on phagocytic cells that recognizes N-formyl peptides, which are released from mitochondria as damage-associated molecular patterns (DAMPs) during tissue injury or by tumor cells. In multiple myeloma, tumor-derived mitochondrial DAMPs activate FPR1 in the bone marrow microenvironment, triggering a pro-inflammatory cytokine response (including IL-6) that facilitates tumor progression. By blocking FPR1, these antibodies aim to suppress this inflammatory niche and reduce tumor burden. Research presented at AACR 2021 demonstrated that blocking FPR1 signaling in syngeneic mouse models of multiple myeloma resulted in significant reduction of tumor expansion, highlighting FPR1 as a potential therapeutic target for hematological malignancies.
