An anti-G-CSF neutralizing antibody is a monoclonal antibody designed to bind to and inhibit the biological activity of Granulocyte Colony-Stimulating Factor (G-CSF, or CSF3). G-CSF is a critical cytokine that regulates the production, differentiation, and mobilization of neutrophils from the bone marrow. While G-CSF itself is a widely used therapeutic for treating neutropenia, neutralizing antibodies against the ligand are primarily employed as research tools to investigate the role of G-CSF in pathological conditions. Preclinical research suggests that blocking G-CSF may be beneficial in treating autoimmune and inflammatory diseases, such as rheumatoid arthritis and neutrophilic dermatoses, as well as in certain cancers where G-CSF-driven myelopoiesis contributes to an immunosuppressive tumor microenvironment. These antibodies function by preventing G-CSF from binding to its cognate receptor (G-CSFR/CSF3R), thereby attenuating neutrophil-mediated inflammation.
