Overview

Anti-GARP refers to a class of therapeutic agents, including monoclonal antibodies and chimeric antigen receptor (CAR) T-cell therapies, that target Glycoprotein A Repetitions Predominant (GARP, also known as LRRC32). GARP is a transmembrane protein primarily expressed on activated regulatory T cells (Tregs), platelets, and certain tumor cells. It functions as a docking receptor for latent transforming growth factor-beta (TGF-β), facilitating its activation and release. By targeting GARP, these therapies aim to inhibit the immunosuppressive effects of TGF-β within the tumor microenvironment, thereby restoring anti-tumor T-cell activity. Some anti-GARP antibodies, such as DS-1055a, also work by depleting GARP-expressing Tregs through antibody-dependent cellular cytotoxicity (ADCC). Several candidates are in clinical development, including Henlius's HLX60 and Daiichi Sankyo's DS-1055a, for the treatment of advanced solid tumors, lymphomas, and gliomas.
