Anti-GARP Chimeric Antigen Receptor-T Cells are a type of investigational cell therapy designed for the treatment of recurrent grade III or IV gliomas. This therapy involves collecting a patient's own T cells and engineering them in the laboratory to express a chimeric antigen receptor (CAR) that specifically targets glycoprotein-A repetitions predominant (GARP), a protein expressed on tumor cells and certain regulatory T cells within the tumor microenvironment. The anti-GARP CAR enables the modified T cells to recognize and attack tumor cells expressing GARP. The therapy is administered via intracavitary infusion directly to the site of the tumor. The primary mechanism is to enhance anti-tumor immunity by disrupting TGF-β signaling mediated by GARP, overcoming immunosuppression and improving tumor response to immunotherapy[1][4][9][10].
