Overview

anti-gD-ds-PBD is an antibody-drug conjugate (ADC) primarily utilized as a non-binding negative control in preclinical oncology research, particularly in studies conducted by Genentech. It consists of a monoclonal antibody directed against the Herpes Simplex Virus (HSV) glycoprotein D (gD)—an antigen that is not expressed on human mammalian cells—conjugated to a potent pyrrolobenzodiazepine (PBD) dimer payload. The "ds" designation refers to the disulfide-based linker used to attach the payload, which is designed to be cleaved within the reducing environment of the intracellular compartment. Because the antibody does not target any human protein, anti-gD-ds-PBD serves as a critical tool for researchers to distinguish between target-mediated therapeutic effects and non-specific toxicity resulting from the systemic release or non-specific uptake of the PBD payload.
