Overview

Anti-GD2 CAR-T cells coexpressing GITRL is an experimental cell therapy designed to treat solid tumors, specifically glioblastoma. It consists of T cells genetically engineered to express a chimeric antigen receptor (CAR) targeting the ganglioside GD2, along with the glucocorticoid-induced TNFR-related protein ligand (GITRL). The co-expression of GITRL is intended to enhance CAR-T cell potency by engaging the GITR receptor, which promotes T-cell activation, reduces exhaustion (lowering PD-1 and LAG-3 expression), and counteracts the immunosuppressive effects of regulatory T cells (Tregs) within the tumor microenvironment. Preclinical studies in glioblastoma models have demonstrated improved functional persistence and metabolic characteristics compared to standard anti-GD2 CAR-T cells.
