Anti-GD2 monoclonal antibody 3F8 is a murine IgG3 monoclonal antibody that targets the ganglioside GD2, which is highly expressed on neuroblastoma cells and has limited distribution in normal tissues. The drug acts primarily through immune-mediated mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). It has been used in combination with granulocyte-macrophage colony-stimulating factor (GM-CSF) as an immunotherapy for high-risk neuroblastoma, particularly for minimal residual disease or refractory cases. Clinical studies have demonstrated its efficacy against neuroblastoma, but the development of human antimouse antibodies can limit repeated dosing due to increased clearance and reduced efficacy. Humanized versions of this antibody have been developed to address immunogenicity concerns[1][5][7].
