The anti-glypican-1 single-chain variable fragment (scFv) is an engineered antibody fragment derived from Miltuximab, a clinical-stage monoclonal antibody developed by GlyTherix that targets glypican-1 (GPC-1). GPC-1 is a heparan sulfate proteoglycan overexpressed in various solid tumors, including glioblastoma (GBM), and serves as a biomarker for targeted therapy and imaging. This scFv consists of the variable regions of the heavy and light chains of Miltuximab connected by a peptide linker. It has been developed and evaluated as an immuno-PET (positron emission tomography) imaging agent, typically radiolabeled with zirconium-89 (89Zr), to enable noninvasive detection of GPC-1-positive tumors. Preclinical studies in GBM models have compared the scFv to the full-length Miltuximab antibody and other fragments (Fab, Fab'2), demonstrating that while the scFv shows high radiolabeling efficiency and receptor-mediated internalization, the full-length antibody provides superior tumor retention and imaging contrast.
