Overview

Anti-glypican-3 chimeric antigen receptor natural killer (CAR-NK) cells are an investigational allogeneic cell therapy developed for the treatment of GPC3-expressing solid tumors. The therapy utilizes natural killer cells or innate lymphoid cells (NK/ILC) derived from HLA-homozygous induced pluripotent stem cells (iPSCs), enabling an "off-the-shelf" manufacturing approach that avoids the logistical complexities of autologous therapies. These cells are genetically engineered to express a chimeric antigen receptor (CAR) specific to glypican-3 (GPC3), a cell-surface oncofetal antigen highly prevalent in hepatocellular carcinoma (HCC), hepatoblastoma, and ovarian clear-cell carcinoma. Upon administration, the CAR-NK cells recognize and bind to GPC3 on tumor cells, triggering targeted cytotoxicity and immune activation. The program emerged from a collaboration between Takeda and Kyoto University's Center for iPS Cell Research and Application (CiRA).
