Overview

Anti-gp100:154-162 T-cell receptor gene-engineered tumor infiltrating lymphocytes is an experimental autologous adoptive cell therapy developed by the National Cancer Institute (NCI) for the treatment of metastatic melanoma. The therapy involves the genetic modification of a patient's own tumor-infiltrating lymphocytes (TIL) or peripheral blood lymphocytes (PBL) using a retroviral vector to express a high-affinity T-cell receptor (TCR). This TCR specifically targets the gp100:154-162 epitope, a melanoma-associated antigen, when presented by the HLA-A*0201 major histocompatibility complex. Administered following lymphodepleting chemotherapy and alongside high-dose aldesleukin (IL-2), these engineered cells are designed to recognize and lyse melanoma cells, offering a potent immunotherapy approach for HLA-A*0201-positive patients.
