Anti-gp49 antibody is a monoclonal antibody directed against the murine immune checkpoint receptor gp49B, which is the ortholog of human leukocyte immunoglobulin-like receptor B4 (LILRB4/ILT3). The antibody functions by blocking the interaction between gp49B and its physiological ligand, fibronectin (specifically the N-terminal 30-kDa module, FN30). This blockade inhibits gp49B-mediated signaling that contributes to class switch recombination in B cells, thereby reducing the secretion of pathogenic IgG autoantibodies. In preclinical studies using the BXSB/Yaa lupus mouse model, administration of anti-gp49 antibodies has demonstrated efficacy in reducing glomerular deposition and ameliorating autoimmune glomerulonephritis.
