Overview

The anti-GPA33 bispecific antibody (specifically huA33-BsAb) is a T-cell-engaging bispecific antibody (T-BsAb) designed for the treatment of GPA33-positive malignancies, such as colorectal and gastric cancers. It is a tetravalent molecule constructed using an IgG(L)-scFv platform, where an anti-CD3 scFv (derived from huOKT3) is fused to the C-terminus of the light chain of a humanized anti-GPA33 antibody (huA33). The antibody functions by simultaneously binding to the glycoprotein A33 (GPA33) on tumor cells and the CD3 epsilon subunit on T cells, thereby redirecting polyclonal cytotoxic T cells to induce tumor cell lysis. Preclinical studies have demonstrated potent T-cell-dependent cellular cytotoxicity (TDCC) in vitro and significant anti-tumor activity in xenograft models. Additionally, anti-GPA33 bispecific antibodies are being investigated in the context of pretargeted radioimmunotherapy (PRIT) using platforms like SADA (Self-Assembling and Dis-Assembling).
