Overview

The anti-GPC1 scFv-Fc fusion protein is a recombinant diagnostic tool designed to detect the expression of glypican-1 (GPC1) in human cancers. Developed by a consortium of Japanese universities, the protein consists of a single-chain variable fragment (scFv) derived from the anti-GPC1 monoclonal antibody clone 1-12, fused to a mouse IgG2a Fc region. This construct is intended to serve as a companion diagnostic or research probe to identify patients suitable for GPC1-targeted CAR-T cell therapies, as the scFv format more closely mimics the binding interface of a CAR-T cell than a full-length antibody. Preclinical studies have demonstrated its ability to successfully detect GPC1 on the cell membrane of lung and esophageal carcinoma cell lines using flow cytometry, fluorescence microscopy, and immunohistochemistry.
