**anti-GPC3-7x19 CAR-T cells** are a novel, autologous chimeric antigen receptor T cell (CAR-T) therapy engineered to target glypican-3 (GPC3), an antigen overexpressed in hepatocellular carcinoma (HCC)[2]. These CAR-T cells are designed with an anti-GPC3 single-chain variable fragment (scFv) fused to CD3ζ, CD28, and 4-1BB co-stimulatory domains. Unlike conventional CAR-Ts, they are further modified to secrete interleukin-7 (IL-7) at moderate levels and chemokine CCL19 at high levels, aiming to re-engineer the tumor microenvironment by recruiting dendritic cells and enhancing T cell persistence and function[2]. Preclinical studies and early clinical experience in HCC patients have shown improved anti-tumor activity and reversal of the immunosuppressive tumor milieu compared to conventional GPC3-CAR-T approaches[2]. Their primary indication is for advanced, GPC3-positive hepatocellular carcinoma[1][2].
